We are investigating the interplay between DNA repair and epigenetics using a stabilized, chemically manipulated version of the drug Decitabine azadC. We will insert the compound into oligonucleotides for biochemical and biophysical studies. Crystal structures with repair proteins and lesion tolerance polymerases as well as detailed repair studies will be performed. Finally we plan to use a systems approach with a newly developed direct sequencing method to learn about the positions where epigenetic changes occur in the genome due to repair and/or blocking of Dnmt enzymes.
